'On' is used for a specific day: Tuesday, Wednesday, 1st June, 2nd June, Christmas Day, Easter Sunday.

She's on the bus - I can't call her.
We're going to Rome on the train.
Everyday he gets on his bike and rides to work.
We use 'on' for all transport which you walk 'on' to. For example, big planes are boarded by walking on, so we say 'I'm on the plane'. Buses are walked 'on' to, so we say 'I'm on the bus'. With a 'bike' you are the object 'on' the bike. Swap the bike for a table and you for keys and it's the same relationship.
